Conditions of Award
- Award funds will be paid directly to the sequencing facility
- When disseminating results, please use the following acknowledgement statement: “This project is part of the MacroMar Project, which is funded by the Moore Foundation. Sequencing was completed at the Bauer Core Facility at Harvard University.”
- Awardees are expected to run the resulting .fastq files through the MacroMar Workflow on MarineOmics to produce a .vcf file and contribute that dataset to the MacroMar project
MacroMar Projects at the Bauer Core Facility
Obtain a Quote
- After receiving the award from MacroMar, we will share your cost estimate with Harvard Bauer Core, who will provide you with two quotes:
- PI quote: A quote for the PI portion (if costs exceed $5200) that you can use to get a purchase order (PO) from your institution.
- MacroMar quote: A quote for the MacroMar portion.
Obtain a PO and upload it to the system
- Create an account using the following link: https://bauercore.fas.harvard.edu/harvard-bauer-core-lockbox-lims
- Unless one of the other options makes sense, select: “Bauer Core Lockbox LIMS External User”
- If your project has the PI covering some costs, obtain a PO from your institution for your amount.
- PO Generation Instructions
- Note: Failure to add the PDF of a valid PO will cause delays to your services
- The PO must be in PDF format and include the company’s logo, company name, and all relevant PO details
- Vendor name and address:
- Harvard University
Central Accounts Receivable
P.O. Box 4999
Boston, MA 02212-4999 USA
- There are multiple Harvard University vendor accounts, make sure to select the correct one listed above
- Screenshots or other documents are not accepted in place of a PDF
- A valid PO must be uploaded to the system before any work can begin at our facilities.
- The core is not responsible for any delays due to billing issues and will not begin work until they are resolved
- Make sure to add “MacroMar Project” on the PO in a “notes” section. If there are any other people you want copied, add that in the notes section above the estimate page during your initial request.
- Create a single PDF with:
- the PI PO
- the MacroMar quote from the first step
- a copy of the spreadsheet estimate turned in with your award.
- You will upload the PDF from step 2 to the system in the next step.
Prepare your samples
- Array samples in full column format in a 96-well PCR plate (semi or full skirted) w/ a foil seal
- Recommended foil plate sealers are VWR #89049-034. If you would like us to mail you some sealers, email k.lotterhos@northeastern.edu
- We recommend that you randomize your samples within and across plates, or at least avoid . This will help to reduce batch effects.
- See the following links for additional requirements
- Submission Details: https://bauercore.fas.harvard.edu/submitting-samples-illumina-sequencing
- DNA Prep Details https://bauercore.fas.harvard.edu/starting-material-dna 1. Harvard will normalize the DNA concentrations before library prep, so you do not have to normalize your samples before you send them. But please follow the prep details and make sure your DNA concentrations are not too high or too low.
- Submit your submission request once you have the PO and sample details https://harb.my.site.com/hbc/s/
- Once your submission has been “Reviewed” in the system, you can send samples to the core
Send your samples
- Ship your samples to the core at this address https://bauercore.fas.harvard.edu/sample-drop.
- If you are in the Boston area, you can also drop them off following the instructions in the link.
- Send an email to nicoleramirez@fas.harvard.edu and illumina_submission-list@lists.fas.harvard.edu to let them know the samples are shipping. Please include “MacroMar Project” on all communications.
Sample processing
- Core staff will “Accept” the request once samples have been successfully received
- Core staff will communicate through the (Laboratory Information Management System) LIMS throughout the process
- After sequencing is completed, you will receive data through the LIMS 1. You are responsible for downloading your data and storing it. If you need assistance or a data storage/processing solution, please contact k.lotterhos@northeastern.edu 2. An email with a copy will be sent to the MacroMar PI (Lotterhos) 3. If there are any other people you want copied, add that in the notes section above the estimate page during your initial request.
- Data Analysis Includes: https://bauercore.fas.harvard.edu/ngs-data-analysis
- Note that for international submission, export of data will go through an export control process which requires the PI Name, Institution Name, and confirmation that this is a Research Based Project
